Brief Introduction:

PNC-27 is a synthetic 32-amino-acid chimeric peptide exclusively for laboratory research, consisting of two functional domains: p53-derived HDM-2 binding fragment and cell-penetrating penetratin sequence. HDM-2 (human MDM2) is highly expressed on the surface of various malignant tumor cells, while almost absent on normal cell membranes. PNC-27 specifically binds to membrane HDM-2, undergoes conformational oligomerization to form transmembrane pores on tumor cell membranes, triggering ion imbalance and cell necrosis independent of p53 function and apoptotic pathways. Part of the peptide penetrates into tumor cells to disrupt mitochondrial membrane integrity, collapse energy metabolism and accelerate tumor cell death. It features ultra-high selectivity, with negligible cytotoxicity to healthy somatic cells, widely applied in oncology pharmacology, tumor targeted lysis, HDM2-p53 signaling and anti-tumor peptide development research.

Main Research Applications:

Tumor targeted killing research: Used in cell and animal models to study selective necrosis of breast, pancreatic, melanoma and leukemia tumor cells.
HDM2 protein interaction research: Suitable for exploring membrane HDM2 binding mechanism and pore-forming pathway.
Mitochondrial tumor damage research: Applied to investigate mitochondrial membrane disruption and energy metabolism collapse in malignant cells.
Differential cell toxicity research: For comparative analysis of safety gap between normal cells and tumor cells.

Novel anticancer peptide screening research: Study selective membranolytic peptide drug design ideas.
